HBTU  is a popular in situ activating reagent commonly used in solid phase peptide synthesis.  It reacts quickly; it is used in rapid Boc protocols utilizing in situ neutralization (M Schnölzer, et al., Int. J. Pept. Protein Res., 1992, 40, 180-93).

 HBTU can react with the exposed peptide N-terminal or unprotected amines in sidechains forming chain-terminating guanidinated by-products (SC Story, JV Aldrich, Int. J. Pept. Protein Res., 1994, 43,292-6) and should not be used in excess of the amino acid component.  In slow or difficult couplings, preactivation of the amino acid with HBTU is recommended. Coupling rates can be enhanced by addition of HOBt. (R Knorr, et al., Tetrahedron Lett., 1989, 30, 1927-30; GE Reid, RG Simpson, Anal. Biochem.1992200, 301-9).
