Fmoc-Amino Acid OPfp Esters HEADING_TITLE

Fmoc amino acid pentafluorophenol esters (Fmoc-OPfp) are activated esters and do not require any additional activation (Kisfaludy L . and Schön I. Synthesis, 1983, 325-7).  In the presence of HOBt, the coupling rate is rapid and few side products are formed.  Since they do not require activation, Fmoc-amino acid pentafluorophenol esters (Fmoc-OPfp esters) are useful in applications where amino acids are partially racemized during coupling (Han Y, Albericio F, Barany G J. Org. Chem. 1997, 62, 4307-12).  Since they do not require activation, Fmoc-Asn-OPfp and Fmoc-Gln-OPfp can be used without formation of cyano side products (Gausepohl H., et al., Int. J. Pept. Protein Res. 1989, 34, 287-94).  Fmoc-amino acid OPfp esters (Fmoc-OPfp esters) were utilized in the homologation of alpha-amino acids to beta-amino acids by the two step Arndt-Eister method (Babu W, et al. J. Pept. Res. 1999, 53, 308-13).  Fmoc-OPfp amino acid esters were also recently utilized in the preparation of positional scanning libraries to determine kinase substrate specificity (Liungdahl T, et al. J. Comb. Chem. 2010, 12, 733-42).
